摘 要:目的 探讨孕激素对体外培养的人早孕蜕膜基质细胞胰岛素样生长因子 - 受体 (IGF- R)基因表达的影响。方法 采用逆转录 -聚合酶链反应 (RT- PCR)技术半定量检测不同浓度孕激素作用 72小时后及 0 .1μm ol/ L孕激素作用不同时间后人早孕蜕膜基质细胞中 IGF- R m RNA的表达水平。结果 体外培养的人早孕蜕膜基质细胞中 IGF- R m RNA呈强阳性表达 ,孕激素降调 IGF- R m RNA的表达 ,并与浓度呈显著负相关 (r=- 0 .6 80 ,P<0 .0 0 1)。孕激素终浓度为 0 .1μm ol/ L时 ,IGF- R m RNA的表达与作用时间无显著相关性 (r=0 .0 0 5 ,P>0 .0 5 )。结论 孕激素可能通过降调人早孕蜕膜基质细胞中 IGF- R m RNA的表达 ,调节基质细胞的增殖和蜕膜化 。Objective To assess the effect of progesterone on the gene expression of insulin-like growth factor-Ⅰ receptor (IGF-ⅠR) in human decidual stromal cells of early pregnancy in vitro. Methods Semiquantitative reverse transcriptase polymerase chain reaction, using β-ACTIN as internal standard,was applied to determine the levels of IGF-ⅠR mRNA in human decidual stromal cells of early pregnancy in vitro after cultured with different concentrations of progesterone for 72 hours or cultured with 0.1μmol/L of progesterone for different periods of time. Results The expression of IGF-ⅠR mRNA was significantly positive in human decidual stromal cells of early pregnancy in vitro.The levels of IGF-ⅠR mRNA were down-regulated by progesterone,and showed significant negative-correlation with the concentration of progesterone (r=-0.680, P<0.001). The levels of IGF-ⅠR mRNA showed no significant correlation with time when the final concentration of progesterone was 0 1 μmol/L (r=0.005,P>0.05). Conclusion Progesterone may play an important role in the regulation of proliferation and decidualization of stromal cells by down-regulating the expression of IGF-ⅠR mRNA in human decidual stromal cells of early pregnancy, which is important for the maintenance of early pregnancy.
