摘 要:目的 :探讨嗜酸细胞 (Eos)凋亡基因Fas抗原的表达及其在凋亡诱导中的作用。方法 :分离正常人外周血Eos ,加入白细胞介素 (IL) - 5与不同浓度 (1- 10 0 0 μg/L)Fas单抗培养 72h ,观察Fas抗原表达、细胞存活和凋亡变化 ,并提取DNA电泳。结果 :新分离的嗜酸细胞表达Fas抗原 ,Eos与单纯介质、IL - 5培养后Fas抗原表达无明显变化。加入不同浓度的Fas单抗对IL - 5介导的Eos存活均有显著抑制作用 ,当Fas单抗浓度为 10 0 0 μg/L时 ,Eos存活率降为 30 %± 12 % (P <0 0 1)。Fas单抗 (10 0 μg/L)处理 ,2 4h后 ,Eos即有明显的凋亡 (35 %± 6 % ,P <0 0 1)。提取DNA电泳呈典型凋亡梯状改变。结论 :Fas抗原参与了Eos凋亡的调节 ,IL - 5抑制Eos凋亡与Fas抗原表达水平无关。AIM: The expression of the Fas Antigen and induction of apoptosis by anti-Fas antibody in esoinophils in vitro were investigated. METHODS: Purified eosinophils from health donors were cultured for 72 h in the presence of human IL-5 and with or without anti-Fas monclonal antibody (MoAb) at various concentrations (1-1000 μg/L). The expression of the Fas antigen on eosinophils was determined by immunocytochemistry. The changes of eosinophils viability and apoptosis were also studied. RESULTS: The Fas antigen was expressed on freshly isolated eosinophils, which had no significant changes after culture in the presence or absence of IL-5. The anti-Fas MoAb at different concentration suppressed significantly the IL-5-mediated eosinophils survival (78%±9%). When eosinophils were cultured in the presence of IL-5 (1×10 4 U/L) with anti-Fas MoAb (1 000 μg/L), the percentage of alive cell decreased to 30%±12%( P< 0.01), and the morphologic changes was consistent with apoptosis. The percentage of apoptosis was significantly increased to 35%±6% after 24 h administration of Fas MoAb ( P< 0.01). DNA extracted from those eosinophils was definitely fragmented. CONCLUSION: The Fas antigen is involved in induction of eosinophil apoptosis, and the level of Fas antigen expression is not definitely related with eosinophils.
