摘  要:目的 研究大黄对烫伤加内毒素“二次打击”后大鼠肝细胞线粒体内单价氧自由基泄漏的影响。方法 采用大鼠背部烫伤模型 ,烫伤 12h后给予内毒素 ( 2 0mg/kg)“二次打击” ,监测肝细胞线粒体内细胞色素氧化酶的活力 ,同时测定肝细胞线粒体内过氧化物歧化酶 (SOD)活力。结果 脓毒症大鼠肝细胞线粒体内细胞色素氧化酶的活力随时间而降低 ,大黄能显著提高细胞色素氧化酶的活力。脓毒症大鼠肝细胞线粒体内SOD大量消耗 ,而大黄能显著降低肝细胞线粒体内SOD的消耗。结论 大黄能显著提高脓毒症大鼠肝细胞线粒体内SOD和细胞色素氧化酶的活力 ,降低肝细胞线粒体内单价泄漏。Objective To investigate the effects of rhubarb on oxygen radicals leakage from the mitochondria of rat liver in sepsis.Methods The burn model was used and endotoxin was administered to the injured rats 12 hrs after the burn.The mitochondria from the liver was isolated.The activity of cytochrome oxidase and superoxide dismutase(SOD) in hepatic mitochondria was measured to evaluate the oxygen radicals leakage from the mitochondria.Results the thermal injury and the second hit induced by endotoxin could progressively decrease the activity of cytochrome oxidase in hepatic mitochondria whereas rhubarb could evidently improve it after the burn and the second hitting.Furthermore,rhubarb could increase the activity of SOD in hepatic mitochondria.Conclusion Rhubarb enhanced the activity of cytochrome oxidase and SOD in hepatic mitochondria in sepsis,which can reduce the leakage of oxygen redicals from the mitochondria.
