摘 要:目的 探索 Sj CTPI和 Sj C2 3DNA疫苗联合应用保护性免疫的协同作用。方法 大量制备 pc DNA3.1- Sj CTPI和 pc DNA3.1- Sj C2 3的质粒 DNA。 4 8只 BAL B/ c小鼠分为 4个组 (A、B、C、D组 )每组 12只。 A组每鼠经双侧股四头肌注射 10 0 μg pc DNA3.1的质粒 DNA,为对照组 ;B组为Sj C2 3组 ,每鼠肌注 10 0μg pc DNA3.1- Sj CTPI的质粒 DNA;C组为 Sj CTPI组 ,每鼠肌注 10 0μgpc DNA3.1- Sj CTPI的质粒 DNA;D组为联合免疫组 ,每组肌注 10 0μg pc DNA3.1- Sj C2 3和 10 0μgpc DNA3.1- Sj CTPI质粒 DNA的混和物。分别于 0、14、2 8d免疫 3次。末次免疫后 30 d,每鼠经腹部皮肤攻击感染 (45± 2 )条日本血吸虫尾蚴。攻击后 4 5 d,剖杀所有小鼠 ,灌冲 ,收集成虫并计数。每鼠肝脏称重后剪碎 ,置 5 % KOH中消化 ,并计数虫卵数。比较各组间的减虫率和减卵率。结果 与对照组相比 ,Sj C2 3组、Sj CTPI及联合组的减虫率分别为 2 8.1%、2 9.1%和 4 1.5 % ,均非常显著地高于对照组 (P分别为 <0 .0 1,<0 .0 1,<0 .0 0 1) ,而联合组则又显著地高于单独 Sj C2 3组及单独 Sj CTPI组 (P均 <0 .0 5 )。三组的减卵率分别为 37.9%、4 4 .2 %和 6 1.4 % ,均非常显著地高于对照组 (P分别为 <0 .0 1,<0 .0 1,<0 .0 0 1) ,但?Objective To investigate the coordination effect on protective immunity by co immunization of SjC23 DNA and SjCTPI DNA vaccines in infected mice of Schistosoma japonicum Methods Large preparation of pcDNA3 1 SjC23 and pcDNA3 1 SjCTPI plasmid DNA. 48 BALB/c mice were divided into four groups (A, B, C, D), 12 for each group. Of group A (Control group), each mouse was injected with 100 μg of pcDNA3 1 plasmid DNA by intramuscle (i.m.). Of group B (SjC23 group), each mouse was injected with 100 μg of pcDNA 3 1 SjC23 plasmid DNA. Of group C (SjCTPI group), each mouse was injected with 100 μg of pcDNA 3 1 SjCTPI plasmid DNA. Of group D (Co immunized group), each mouse was injected with the mixture of 100 μg pcDNA3 1 SjC23 and 100 μg pcDNA 3 1 SjCTPI plasmid DNA. At day 0, 14, 28, each mouse was immunized with the same dosage and same method. At day 30 after last immunization, each mouse was challenged with 45±2 cercariae of Schistosoma japonicum Chinese strain. At day 45 after challenge the mice were sacrificed and perfused, the number of recovered adult worm and hepatic eggs were counted. The worm reduction rate and egg reduction rate were compared between groups. Results The worm reduction rates in SjC23 group (B), SjCTPI group (C) and co immunized group (D) were 28 1%, 29 1% and 41 5% compared with control group respectively. They were significantly higher than that in control group (P<0 01, <0 01, <0 001). Furthermore, the worm reduction rate in co immunized group was higher than that in SjC23 group or in SjCTPI group(P<0 05, <0 05). The egg reduction rates in B, C, D groups were 37 9%, 44 2% and 61 4% compared with control group respectively. They were also significantly higher than that in control group (P <0 01, <0 01, <0 001). The egg reduction rate in group D was higher than that in group B or in group C (P<0 05,<0 05). Conclusion Co immunization with SjC23 DNA and SjCTPI DNA vaccines could significantly enhance the protective effect compared with only SjC23 DNA
