摘  要:本文用甲状腺素(T_4)甲酯—BSA抗原免疫BALB/C小鼠,获得4株分泌抗人甲状腺素单克隆抗体的杂交瘤细胞系。亚类鉴定均属IgG_1型。将单克隆抗体应用在放射免疫分析(RIA)中,取代多克隆抗体,证实其在甲状腺疾病诊断中,甲状腺机能亢进的诊断符合率高于多克隆抗体,显示其特异性强、灵敏度高、亲和力好(亲和常数K=1.26x10^(10)L/M)、反应平衡时间短等优点。Thyroxine(T_4)was firstly methylesterized and coupled to bovine serum albumin (BSA) to form T_4-methylesterBSA. Then BALB/C mice were immunizcd. The spleen cells of mice were fused with mouse FO myeloma cells. Then the monoclonal antibodies were prepared by selective antibody-producing clones. Subclass determination showed that these MoAbs belong to IgG1. The McAbs has the advantages of high sensitivity, specifity and affinity (K=1.26×10^(10)L/M). Using these MoAbs in T_4 radioimmunassay, the time of reaction to equilibrium was only 15-30 minutes. The preliminary clinical application showed that the diagnostic agreement rate to hyperthyroidism of using MoAbs was higher than that of using rabhit antiserum, the former was 92.1%, while the later was 84.8%. The diagnosis agreement rate to hypothroidism was 100%. The lowest determinable value was 1.1μg/L.
