摘  要:通过氯化铈 (Ce Cl3)沉淀的电镜细胞化学法 ,对比观察了抗寒冬小麦幼苗和冷敏感春小麦幼苗质膜 Ca2 + -ATPase活性的变化 ,结果显示 :2 0℃下生长的冬小麦幼苗 ,Ca2 + - ATPase活性主要定位于质膜上。2℃下 ,随处理时间的延长 (3h、12 h) ,质膜 Ca2 + - ATPase活性反应逐步增强。2℃下处理 3d,质膜上仍存在酶反应产物 ,质膜 Ca2 + - ATPase活性高于对照。2 0℃下生长的春小麦幼苗 ,其质膜 Ca2 + - ATPase活性水平高于同样条件处理下的冬小麦幼苗的酶活性水平。2℃处理 3h,春小麦幼苗质膜上酶活性反应明显增强。2℃处理 12 h,酶活性反应迅速降低。2℃下处理 3d,春小麦幼苗质膜上酶反应产物明显少于对照 ,质膜 Ca2 + - ATPase已接近失活。结果表明 ,低温逆境下质膜 Ca2 + - ATPase活性的大小及其稳定性 。Using cerium trichloride precipitation method with electromicroscopic cytochemical assay, the difference of Ca 2+ ATPase activity in plasmolemma between seedlings of cold resistant winter wheat and cold sensitive spring wheat was compared. Grown under 20℃, the Ca 2+ ATPase activity of winter wheat seedlings was mainly localized in plasmolemma. Treated at 2℃ for a period of times (3 h? 12 h), the Ca 2+ ATPase activity increased gradually, and when the same treatment lasted for 3d, the enzyme activity still remained active and was higher than the control. For spring wheat seedlings grown under 20℃, the level of Ca 2+ ATPase activity in plasmolemma was similar with that of winter wheat seedlings in the same treatment. When treated at 2℃for 3 h, the enzyme activity in plasmolemma of spring wheat seedlings increased considerably, and when treated at 2℃ for 12 h, a sharp decrease in the enzyme activity was observed and the product of the enzyme activity was less than that of winter wheat seedlings. When the treatment lasted for 3 d, the product of the enzyme activity was less than the control and the enzyme was almost inactivated. It is concluded that activity and stability of Ca 2+ ATPase under 2℃low temperature are the key factors in the development of cold resistance of winter wheat.
