摘 要:环境扫描电镜可以对很多含水的、导电性差的生物样品进行直接观察 ,从而避免用常规扫描电镜观察时 ,由于生物样品经脱水、临界点干燥和导电处理而可能引起的人为变化。本文利用环境扫描电镜 ,对几种热带水果荔枝、龙眼、芒果、木瓜、杨桃的花粉及其相应的器官进行了直接观察 。Most biological samples contain water higher than 85% with bad electric conductivity. When observed by normal scanning electron microscope under high vacuum, the samples need to be dehydrated, critical point dried and electrical conduct treated. This series of treatments will result artificial changes in structure. The environmental scanning electron microscope can be utilized to directly observe the biological samples with high content of water. It will avoid the artificial changes probably resulted from the samples treated through dehydration, critical point dry, and electrical conduction. By using environmental scanning electron microscope, the pollen and its related organs from some tropical fruits including Litchi, Longan, Mangifera, Carica papaya, and carambola were observed, and the results were reported. Our results demonstrated that the biological samples can be directly observed using environmental scanning electron microscope without pre treatments.
