摘 要:介绍一种定量观察血管内皮细胞表面阴离子场的技术方法 ,以研究毛细血管腔面阴离子场与血管通透性的关系。用前包埋程序以阳离子胶体金 (CCG)做为探针 ,标记硬脑膜毛细血管内皮腔面的阴离子场 ,在电子显微镜下摄影计数内皮细胞表面标记的金颗粒。在硬脑膜毛细血管腔面 ,微绒毛和质膜小泡的表面均见 CCG标记 ,而不带电荷的 BSA -胶体金呈阴性结果。前包埋阳离子胶体金标记技术可特异性地显示硬脑膜血管内皮细胞表面的阴离子场 ,并可做定量研究。标记微环境 ,血管开口大小和复染可能影响标记结果。Object: A special technique for quantifying the anionic sites of vascular endothelial cells is introduced so as to study the relationship between the capillary vessel anionic sites and vascular permeability. Methods: Dural capillary vessel surface anionic sites were labelled using cationic colloidal gold as probe following the pre embedding program and the gold particles on the surface of vascular endothelial cells were counted by electron microscope photography. Results: CCG labelling particles were seen on the surface of the lumen, the microvilli and plasmalemmal vesicles of the dural capillary vessel, while the non electrified BSA CG labelling particles presented negative result. Conclusion: Pre embedding cationic colloidal gold (CCG) labelling technique can reveal the anionic sites on the surface of dural vascular endothelial cells quantitatively. Labelling microcircumstance, the size of vessel opening diameter and counterstain may also affect the result.
