银屑病皮损中IL-10及IL-12p35、p40mRNA表达的研究

摘　　要：探讨 IL- 10和 IL- 12在银屑病发病机理中的可能作用 ,为应用基因治疗银屑病提供理论基础。采用逆转录聚合酶链反应 (RT- PCR)技术检测了 12例银屑病患者及 6例正常皮肤组织中 IL- 10和 IL- 12 p35、p40 m RNA的表达情况。研究结果表明 :(1)与正常皮肤组织相比银屑病皮损中 IL- 10 m RNA明显降低 (P<0 .0 1)。 (2 ) IL- 12 p35 m RNA在银屑病皮损和正常皮肤组织中均呈阳性表达 ,IL- 12 p40 m RNA只在银屑病皮损中呈阳性表达 ,而在正常皮肤中为阴性。推测 IL- 12在银屑病的发生和发展中可能起重要作用 ,而 IL- 10可能在银屑病消退中起一定作用。To investigate the possible role of IL-10 and IL-12 in the pathogenesis so as to supply theoretical basis for the gene therapy of psoriasis,the expressions of IL-10 and IL-12 p35、p40 mRNA in the 12 cases of psoriatic lesions and 6 normal skin tissues were studied with RT-PCR technique The results showed that the expressions of IL-10 mRNA in the psoriatic lesions were significantly lower than those in the normal skin tissues ( P <0 01); the expressions of IL-12 p35 mRNA were positive both in the psoriatic lesions and in the normal skin tissues; IL-12 p40 mRNA were positively expression only in the psoriatic lesions but negative in the normal skin tissues It was suggested that IL-12 might play an important role in the occurrence and progression of psoriasis, while IL-10 might have certain role in the regression of psoriasis
