摘  要:用胰岛素(INS)免疫豚鼠制得INS抗血清,通过氯胺T法碘化标记INS制备^(125)I-INS,结合本室研制的可磁化固相二抗分离试剂(DAMP),建立了血清INS可磁化固相放射免疫分析法(RIA)。本法灵敏度达0.1μU,回收率>95%,批内变异(CV)<2.3%,批间CV<7.4%。并具有不需离心,操作简便,分离迅速的特点。经临床初步应用,获得满意的结果。A new radioimmunoassay for insulin in serum with magnetic separation of the solid-phase is described. The insulin antiserum was obtained from immunizing guinea pig with insulin. 125I-insulin was labeled by use of the io-dinating reagent chloramine-T. The double antibody magnetizable part ides (DAMP) were developed by our laboratory. The lowest detection limit was 0.1 μU. The recovery of the method was more than 95%. The coefficients of variation within assay were less than 2.3% while those between assays were less than 7.4%. This method avoids the need for centrifugation, provides a simple, rapid and universal separation procedure, and has been applied clinically with satisfactory results.
