抗约氏疟原虫感染过程中单核-巨噬细胞免疫效应的研究

摘  要:目的 探讨单核 -巨噬细胞及其分泌的NO在抗疟原虫感染过程中的作用。方法 应用吉姆萨薄血膜染色法和腹腔注射鸡红细胞法分别检测红细胞感染率及单核细胞和腹腔MΦ的吞噬能力 ;通过Griess反应检测小鼠腹腔MΦ合成NO水平。结果 腹腔MΦ在感染后 6d吞噬率即达到高峰 ,单核细胞的数目和吞噬能力在感染 6d后开始增加 ,同时 ,虫体血症水平升高的趋势受到遏制 ;感染早期MΦ合成NO水平逐渐升高 ,但对虫体血症没有影响。结论 单核 -巨噬细胞数量的增加和巨大吞噬杀伤作用的发挥可削减并最终清除虫体血症 ;Aim To investigate the immunologic effects of mononuclear macrophages and NO synthesized by mononuclear macrophages on parasites during the infection of Plasmodium yoelii.Method With Giemsa staining and injecting chicken RBC to abdominis of mice,the phagocytic function of monocytes and peritoneal macrophages were observed;the NO levels in cultured supernatants from peritoneal macrophages of infected mice were detected with Griess reaction.At same time,the effects of both on the parasitemia were examined.Results The phagocytic rate of peritoneal macrophages reached the highest peak at 6 th day after infection,the quantity and phagocytic power of monocytes started to increase at 6 th day after infection and simultaneously the increasing trend of parasitemia was inhibited;the NO levels synthesized by peritoneal macrophages elevated gradually during the early stage of infection,but it did not affect the changes of parasitemia.Conclusion The increase of mononuclear macrophages in quantity and phagocytic power can put down and eliminate parasitemia eventually;but at the early stage of infection the meaning of NO increase synthesized by peritoneal macrophages need further research.
