鱼棉平喘方对哮喘豚鼠血浆中cAMP、cGMP及PAF含量影响

摘　　要：目的 :探讨鱼棉平喘方 (DYA)对嗜喘豚鼠的平喘作用及作用机理。方法采用氯化乙酰胆碱和磷酸组胺混合液雾化引喘 ,观察豚鼠引喘潜伏期的变化 ;采用 4 %卵清蛋白生理盐水和 4 %氢氧化铝凝胶致敏 ,卵清蛋白复制哮喘模型 ,生物定量法测定血浆中血小板活化因子 (PAF)的含量。结果 :DYA给药后各组豚鼠的引喘潜伏期比给药前及正常组均显著延长 (P <0 .0 1 )。DYA治疗各组与模型组比较 ,血浆cAMP含量及cAMP/cGMP比值均显著升高 (P <0 .0 1 ) ,血浆cGMP含量显著降低 (P <0 .0 1 )。结论 :DYA有明显的平喘作用 ,其作用机理与其调节机体cAMP、cGMP水平 ,稳定cAMP/cGMP比值 。Objective: To investigate the anti asthmatic effects and its mechanism of Yumian Anti asthma Formula (DYA) on asthmatic guinea pigs. Methods: Observe the change of the latent period of asthma inducing of normal guinea pigs and that of them after DYA preventing. Determine the plasma cAMP and cGMP contents. Results: The latent period of asthma inducing was obviously prolonged in guinea pigs prevented by DYA when compared with that of normal guinea pigs and guinea pigs before given DYA ( P <0.01). The plasma cAMP and the ratio of cAMP/cGMP increased obviously but cGMP level decreased in DYA treated asthmatic guinea pigs when compared with those of untreated asthmatic guinea pigs ( P <0.01～0.05). The plasma PAF contents of DYA treated asthmatic guinea pigs were much lower than those of asthmatic guinea pigs ( P <0.01), but still much higher than those of normal guinea pigs ( P <0.01). Conclusion: DYA has obvious effects of relieving asthma on asthmatic guinea pigs and its mechanism is due to the regulation of the plasma cAMP, cGMP and PAF level and the stabilization of the ratio of cAMP/cGMP.
