摘　　要：目的 :探讨基因组DNA的制备方法。方法 :酚 -氯仿法、改良盐析法及口腔粘膜法。结果 :三种方法提取的DNA产量均能满足PCR及基因分型的需要。结论 :改良的盐析法相对简单、经济和快速 ,更适合临床上基因诊断的需要 ;而口腔粘膜法特别适用于儿童及家系调查。Objective:To determine the preparation methods of genomic DNA. Methods:We used three different ways. They are Hydroxybezne-chloroform method and improved salt out method by which genomic DNA can be isolated from human blood samples and mouth mucous membrane method by which genomic DNA was extracted from buccomucosal epithelial cells. All of the yields and qualities of the genomic DNA gained by the three ways were compared. Results:Every yield and quality of genomic DNA met the demands of PCR and genotyping. Conclusions:Improved salt out method is a comparatively simple, economical and rapid way, and it is more suitable for clinical genetic diagnosis, while mouth mucocous membrane method is specially suitable for young children and genealogical researches.
