软肝化癥丸对肝星状细胞活化的影响

摘　　要：观察软肝化疒徵丸干预肝纤维化的主要作用机制。分离培养大鼠肝星状细胞 (HSC) ,以软肝化疒徵 丸灌胃正常大鼠 ,制备药物血清 ,温育培养细胞。 3H TdR掺入法测定细胞增殖 ,ELISA法测定细胞上清Ⅰ型胶原含量 ,RT PCR法观察前胶原α2 (Ⅰ )mRNA的表达 ,同位素标记法测定细胞上清间质胶原酶的活性。结果 :软肝化疒徵丸药物血清能抑制HSC的增殖与胶原蛋白的分泌 ,提高其间质胶原酶的活性 ,抑制其α2 (Ⅰ )mRNA的表达。结论 :软肝化疒徵The main mechanism of the interfering effects of Ruanganhuazheng Pills (RP) on hepatic fibrosis was studied. Rat hepatic satellite cells (HSC) were separated and cultured with RP serum prepared in normal rats. JH3,D,Z TdR incorporation was used for detecting HSC proliferation; ELISA, for measuring the level of collagen I in HSC supernatant; RT PCR, for detecting the expression of mRNA of HSC procollagen α 2 (I) and, the isotope label method, for detecting the activity of MMP1 in HSC supernatant. The results showed that RP-serum could inhibit HSC proliferation and the secretion of collagen I, increase the activity of MMP1, decrease the expression of mRNA of procollagen α 2 (I), which implies that the mechanism of the effects of RP for interfering hepatic fibrosis may be its inhibiting the activation of HSC.
