摘  要:应用cDNA AFLP(Amplifiedfragmentlengthpolymorphism)技术 ,分析了棉花洞A雄性不育和可育材料在花药发育过程中花粉发育相关基因表达的差异。结果表明 :在花药发育的相同时期 ,不育和可育花粉cDNA AFLP的谱带在单核期的差异大于减数分裂期。在花粉发育过程中 ,花粉发育相关基因的表达具有时空特异性。共回收了 6 4条差异cDNA片段 ,随机选择 3个片段标记为探针 ,RNA斑点杂交分析表明 ,GHA2 7具有花器官组织特异性 ,仅在花器官中表达 ;GHA2 8、GHA4 7则具有花药组织特异性且仅在可育花药中表达。序列相似性比较表明 :GHA2 7与植物ADP ribosylationfactor (ARF)基因有较高的相似性 ,而GHA2 8、GHA4cDNA AFLP, an effective method for mRNA differential display, was employed to compare the gene expression in developing anthers between the male sterile and fertile plants of cotton (Gossypium hirsutum L.)cultivar, Dong A. In the micro spore stage, there were more differential bands of cDNA AFLP than that in the meio phase stage. Among 64 differential fragments produced by cDNA AFLP, three were randomly selected for further analysis. RNA dot blotting showed that the GHA27 transcript was expressed mainly in floral tissues; on the other hand, the GHA28 and GHA47 transcripts were present specifically in anther. BLAST analysis demonstrated that GHA27 was highly similar to the plant ADP ribosylation factor genes, while GHA28 and GHA27 were shown no significant similar to any sequences in the available databases.
