芽殖和/或附属物细菌群中的一个新属

摘　　要：从我国江苏省如东县掘东垦区大米草根内分离到一株氧化型革兰氏阴性细菌,编号4088。该菌株具有两种出芽繁殖方式:子细胞主要从母细胞顶端长出柄状附属物,这种附属物末端膨大形成子细胞和直接从母细胞上生长出子细胞(约占20%)。细胞内有聚β-羟基丁酸盐颗粒。细胞外有荚膜。菌落产生β-葫萝卜素黄色素。呼吸型代谢,从不发酵。好氧生长,无氮情况下行微好氧生长。能沉淀铁锰。能固定大气中的氮。有机化能营养。从糖类和醇类产碱而不产酸。不利用甲醇、甲酸为唯一碳源。DNA 中 G+C 含量为70.1—70.4mol%。根据以上特性4088菌株与芽殖和/或附属物细菌群中的某些属相近,但出芽繁殖方式不同于已知属。因此,将4088菌株归属于芽殖和/或附属物细菌群中的一个新属——双殖菌属(Dipareforma gen.nov.Zhou et al.),模式种为米草双殖菌(Dipareforma spartinacae sp.nov.Zhou et al.)。菌种保藏于中国科学院微生物研究所菌种保藏室。菌株编号为 AS1.1762。A strain of oxidative Gram negative ba- cteria,No.4088,was isolated from the root of spartina anglica grown at the beach of Rudong County,Jiangsu Province.The cells of the strain No.4088 are rod or ovoid shaped,0.8— 1.0 by 1.1—1.5μm,with capsule,non-motile. The granules of poly-β-hydroxybutyrate and yellow pigment of β-carotin type are produced. Strain No.4088 multiplies with two forms of budding.The one is buds formed primarily by swelling of the prosthecae (hyphae).The other is with yeast-like form;about 20% buds sprout from mother cells directly.These two types of budding may appear on the same cell Strain No.4088 is an organism with respiratory metabolism,never fermentative,aerobic,mic- roaerophilic under combined nitrogen-free con- dition.Chemoorganotrophic,no acid is pro- duced from sugars and alcohols.Glucose, xylose,maltose,acetate,citrate,succinate, malate,α-ketoglutarate,malonate,arginine and casein are utilized as the sole carbon and en- ergy sources.Methanol,formate,propionate, butyrate,iso-butyrate,valerate,iso-valerate,ca- prate,fumarate,huppurate,aminoacetate,pro- line,2,3-butylene glycol,or betaine is not utili- zed.Oxidase and catalase are positive. MR and VP are negative,Hydrogen sul- fide is not produced.Nitrate is redu- ced,Litmus milk becomes alkaline.Escu- lin,starch or gelatin is not hydrolized. Lysine decarboxylase and arginine decar- boxylase are positive and ornithine decarbo- xylase,DNase and lipase,urease,arginine dihydrolase,phenylalanine deaminase are nega- tive.Free nitrogen can be fixed.Iron and manganese can be deposited.Growth occurs in medium containing NaCl not more than 1.5%.Growth temperature:20—35℃,30℃ is optimum.Growth pH 6.0—9.0,optimum is pH 7.5.Guanine and Cytidine contents in DNA is 70.1—70.4 mol%.
