种鸡场鸡白血病净化的研究(Ⅱ)——鸡白血病病毒ELISA检测抗体包被稳定剂的研究

摘　　要：本研究应用双抗体夹心酶联免疫吸附试验 (DAS- EL ISA) ,对吸附在酶标板上的抗体 (兔抗禽白血病病毒群特异性抗原的 Ig G)通过 4种稳定剂处理之后 ,置于 4～ 8℃、室温 (18～ 2 3℃ )、37℃条件下 ,存放 11天、19天 ,结果发现 :利用 DAS-EL ISA检测同一 AL V阳性蛋清样品 ,测定 OD值大小不同。选择合适的稳定剂处理酶标板 ,在 4～ 8℃条件下保存半年 ,对吸附在酶标板上抗体活性进行探讨 ,其中以山梨醇和 PVP为代表的两种稳定剂效果良好 ,和常规包被抗体 (4～ 8℃隔夜 )The study was based on the principle of DAS ELISA.The plates coated by the antibody(rabbit IgG against gs antigen of avian leukosis virus)were dealt with four stabilizers, then separately stored at 4～8℃,at room temperature (18～23℃),and at 37℃ for 11 days and 19 days.The result showed that the OD value of the same positive sample from egg albumen were different by DAS ELISA.While the plates coated by the antibody were handled by proper stabilizers,stored at 4～8℃ for half year,the antibody's reactivity did not significantly differ from the plates which were treated normally in detecting the same ALV positive sample.Among the stabilizers,those with sorbitol and PVP was the best.
