雷公藤红素对狼疮鼠肾组织Ⅲ型胶原和层粘素的影响

摘　　要：目的 :研究雷公藤红素 (Tripterine)对狼疮小鼠肾小球硬化的防治作用。　 方法 :在不同发病时间、用不同剂量的雷公藤红素对BWF1小鼠进行腹腔注射。每 2周测鼠 2 4h尿蛋白量及血清抗dsDNA抗体水平。 2 0周后检测鼠肾组织Ⅲ型胶原 (collagentypeⅢ ,ColⅢ )、层粘素 (laminin ,LM)、基质金属蛋白酶 (MMP) 1及其组织抑制剂 (TIMP) 1、转化生长因子 (transforminggrowthfactor,TGF) β1mRNA的表达。　 结果 :①雷公藤红素抑制小鼠蛋白尿及血清抗dsDNA抗体水平 ,增加肾局部MMP 1而减少ColⅢ、LM、TIMP 1及TGF β1mRNA的表达 ,改善鼠肾组织学病变。②蛋白尿发生前使用雷公藤红素较蛋白尿发生后使用疗效更显著。③蛋白尿发生前使用雷公藤红素 3mg/ (kg·wk)和 6mg/ (kg·wk)的疗效无统计学差异。　 结论 :雷公藤红素对该狼疮模型的肾小球硬化具有保护作用。其降低肾脏ColⅢ及LM沉积可能是通过增加小鼠肾局部MMP 1而抑制TGF β1及TIMPObjective:To study the renal protective effects of Tripterine in the lupus nephritis mice. Methodology:53 non proteinuria BW F1 mice were divided randomly into 4 groups.They were control group A,the tripterine treated group 1B,1C and 2B.3mg and 6 mg/(kg·wk) of tripterine were injected peritoneally to the mice of group 1B and 1C separately at the beginning study.6 weeks later,6 mg/(kg·wk) of tripterine were given to the mice of group 2B when they developed proteinuria.The levels of urinary protein excretion in 24 hour and serum anti ds DNA antibody were measured every two weeks.The local expressions of renal collage type Ⅲ,laminin,MMP 1,TIMP 1 were analyzed with immunohistochemistry.RT nested PCR was used to evaluate the expressions of renal TGF β1 at the end of this study. Results:①Tripterine suppressed the development of proteinuria,decreased the level of serum anti ds DNA antibody,reduced the expressions of TGF β1 mRNA,collagen type Ⅲ,laminin,TIMP 1 and improved the expression of MMP 1 in kidney of murine lupus.②There was greater protective effect of tripterine before occurrence of proteinuria compared with that after the occurrence of proteinuria in lupus mice.③No significance of the proteinuria protective effect was found between the 3mg and 6 mg/(kg·wk) of tripterine treated groups. Conclusion:Tripterine has definite renal protective effects in the lupus murine model.The decrease of renal collagen type Ⅲ and laminin is probably due to its suppressive effects on the local expressions of TGF β1,TIMP 1 and its improvement effect on the local expression of MMP 1.
