尖锐湿疣患者PBLC Fas、Bcl-2的表达与血清IFN-γ、IL-4的相关性研究

摘　　要：目的　研究尖锐湿疣 (CA)患者外周血淋巴细胞 (PBLC)凋亡调控蛋白表达和血清IFN γ、IL 4水平的相关性。方法　采用流式细胞仪的间接免疫荧光素标记法和双抗体夹心ELISA法 ,分别检测了 60例CA患者PBLC凋亡调控蛋白Fas、Bcl 2的表达水平和血清IFN γ、IL 4水平。结果　CA患者PBLC的Fas表达水平明显高于健康对照组 ,Bcl 2则明显低于健康对照组 (P均 <0 .0 1) ;CA患者血清IFN γ水平明显低于健康对照组 ,IL 4水平明显高于健康对照组 (P均 <0 .0 1) ;CA患者PBLCFas表达水平与血清IFN γ水平呈负相关 ,而与血清IL 4水平呈正相关 ;Bcl 2表达水平与血清IFN γ水平呈正相关 ,而与血清IL 4水平呈负相关。相关系数经t检验差异均有极显著性 (P <0 .0 1)。结论　CA患者PBLC有凋亡异常 ,凋亡异常与TH1型细胞因子分泌低下及TH2 型细胞因子分泌增加之间有一定的相关性 ,在CA的免疫发病机制中可能起重要作用。Objective To investigate the correlation of the expression of the apoptotic regulatory proteins in public blood lymphocytes(PBLC) and the level of IFN γ,IL 4 in serum of patients with condyloma acuminata(CA).Methods Indirect immunofluorescein labeling method of flow cytometer and solid sandwich Enzyme Linked Immunoabsorbent assay(ELISA) were performed for detecting the expression of the control protein of apoptosis Fas,Bcl 2 in PBLC and the level of IFN γ,IL 4 in serum of 60 cases of CA.Results The expressions of Fas in PBLC of CA was significantly higher than that of the normal control,but Bcl 2 was significantly lower(both P <0.01);the level of IFN γ in serum of CA is significantly lower than that of normal control,but IL 4 was significantly higher(both P <0.01),the expression of Fas in PBLC had a negative correlation with the level of IFN γ in serum of patients with CA,but had a positive correlation with the level of IL 4,the expression of Bcl 2 had a positive correlation with the level of IFN γ,but had a negative correlation with the level of IL 4.All the correlation coefficients had significance by t test( P <0.01).Conclusion Patients with CA were characterized by the abnormal apoptosis in PBLC and the suppressed secretion of the TH 1 associated cytokines(eg:IFN γ),the increased secretion of the TH 2 associated cytokines(eg:IL 4),between these two aspects may have a correlation and may play an important role in the immune pathogenesis of CA.
