摘  要:目的 主要观察母鼠经口给予谷氨酸单钠 ig对仔鼠大脑皮层和海马神经元 Bax和 Bcl- 2表达的改变 ,探讨谷氨酸的兴奋毒性作用 ;并观察鼠大脑皮层和海马神经元 Bax的改变与 Bcl- 2表达的改变是否有互动关系 .方法 妊娠 17~2 1d的昆明系小鼠经口 ig,给予谷氨酸单钠 (4 mg· g- 1 ) ,仔鼠产后第 10 ,2 0和 30日采用免疫组织化学方法检测了 Bax/Bcl- 2在鼠脑皮层和海马组织中的表达 .细胞损伤采用镜下细胞直接计数法 .结果 胚胎期摄入谷氨酸可引起 10 ,2 0和30日龄仔鼠大脑皮层、海马 CA1,CA2 ,CA3和 CA4区神经元 Bax表达的增高 (d10 . 36 .0 vs16 .6 ,47.0 vs2 6 .0 ,74.0vs2 5 .0 ,34 .3vs2 1.3,42 .6 vs2 2 .0 ,P<0 .0 5 ;d2 0 . 2 7.0 vs2 0 .0 ,31.0 vs 9.7,5 2 .3vs 17.0 ,2 6 .5 vs 10 .0 ,6 3.7vs2 2 .0 ,P <0 .0 5 ;d30 . 2 5 .0 vs 9.7,32 .0 vs 10 .0 ,38.3vs14.7,2 6 .0 vs10 .0 ,31.7vs9.7,P<0 .0 5 ,cells· 0 .0 6 33mm- 2 ) ,而相应地 ,10 ,2 0日龄仔鼠脑皮层和海马 CA1,CA2 ,CA3和 CA4区神经元 Bcl- 2的表达比正常鼠明显降低(d10 . 13.7vs44 .7,12 .0 vs37.7,2 3.0 vs6 1.7,11.0 vs31.0 ,18.7vs5 1.7,P<0 .0 5 ;d2 0 . 14.0 vs36 .7,14.0 vs32 .7,19.0 vs 43.7,10 .3vs 2 7.3,19.3vsAIM This study aimed at exploring excitotoxicity of monosodium glutamate (MSG) in the offspring cerebral cortex and hippocampal subregions after maternal MSG oral administration, and whether a change in Bax content was associated with a response in Bcl 2 expression. METHODS Kunming mice were given per os MSG (4 mg·g -1 body weight) at 17~21 d of pregnancy and their offsprings′ behavior was studied on postnatal days of 10, 20 and 30. By using immunohistochemical analysis, we examined the involvement of Bcl 2 and Bax in the glutamate induced cell death in cortical and hippocampal neurons. Cell damage was assessed by direct cell counting. RESULTS Administration of MSG during the fetal period in mice resulted in a moderate increase in the expression of Bax in principal neurons in the cerebral cortex, CA1, CA2, CA3, and CA4 on postnatal days of 10, 20 and 30 in the offspring mice (d10. 36.0 vs 16.6 , 47.0 vs 26.0 , 74.0 vs 25.0, 34.3 vs 21.3, 42.6 vs 22.0 , P < 0.05 ; d20. 27.0 vs 20.0, 31.0 vs 9.7 , 52.3 vs 17.0, 26.5 vs 10.0 , 63.7 vs 22.0, P <0.05;d30. 25.0 vs 9.7 , 32.0 vs 10.0, 38.3 vs 14.7, 26.0 vs 10.0, 31.7 vs 9.7, P <0.05, cells·0.0633 mm -2 ). In contrast, Bcl 2 protein expression was lowered significantly by glutamate exposure in principal neurons in the cerebral cortex, CA1, CA2, CA3, and CA4 on postnatal days of 10, 20 compared with that of the control (d10. 13.7 vs 44.7, 12.0 vs 37.7, 23.0 vs 61.7, 11.0 vs 31.0 , 18.7 vs 51.7, P <0.05;d20. 14.0 vs 36.7, 14.0 vs 32.7 , 19.0 vs 43.7, 10.3 vs 27.3, 19.3 vs 34.7, P <0.05, cells·0.0633 mm -2 ). Bcl 2 protein expression was also lowered significantly by glutamate exposure in principal neurons in the cerebral cortex on postnatal days of 30 (23.3 vs 33.0, P <0.05, cells·0.0633 mm -2 ).
