摘 要:目的 探讨老龄大鼠大脑皮质基因组DNA中 8 羟基 2′ 脱氧鸟苷 (8 OH dG)变化规律和左归丸的调节作用。方法 建立 2 4月龄自然衰老实验模型 ,随机分为老年组 (生理盐水灌胃 )和治疗组 (左归丸灌胃 ) ,处理 3个月 ;以 3月龄大鼠为青年对照组。取大脑皮质 ,抽提DNA ,采用酶联免疫吸附法 (ELISA)测定 8 OH dG水平。结果 老龄大鼠大脑皮质基因组DNA 8 OH dG水平比青年对照组明显升高 ,左归丸组低于老年组 (P <0 .0 5)。结论 8 OH dG在大脑皮质老化过程中上调 。Purpose: To explore the altered concentrations of 8-hydroxy-2′ -deoxyguanosine (8-OH-dG) in cerebral genome of aging rats and regulative effect of Zuogui Wan(ZGW) on it. Methods Aging rat models were established by being fed for 24 months and randomly divided into two groups, aging group and ZGW group by three-month administration of 0.9% NS and ZGW liquid respectively, and controlled by young group which consisted of three-month-old rats. Cerebral samples were harvested from rat brain of every group and the levels of 8-OH-dG was assessed by enzyme-linked immunosorbent assay (ELISA). Results: The levels of 8-OH-dG in cerebral genome of aging rats were higher than the young control while decreased after ZGW administration. Conclusions: It is suggested that the upregulated contents of 8-OH-dG in genome of aging neocortex may be related to agravating brain senescence and contributing to neural degenerative desease. ZGW has effect of neural protection and repairment on brain through down-regulating genomic 8-OH-dG.
