早孕期蜕膜组织细胞凋亡异常对妊娠的影响

摘　　要：目的探讨早孕期蜕膜组织细胞凋亡异常与妊娠维持的关系。方法应用TUNEL法检测凋亡的发生,原位杂交和免疫组化法检测Fas、FasL的表达。结果(1)正常早孕41d蜕膜组织细胞大量凋亡,Fas、FasL的mRNA及蛋白表达较强;(2)正常早孕54d凋亡细胞明显减少,Fas、FasL的表达明显减弱;(3)早孕55d自然流产组,蜕膜组织大量凋亡,且Fas、FasL的表达较正常早孕54d显著增强(P<0.05)。结论早孕期蜕膜组织凋亡异常可能是自然流产的机制之一,Fas途径可能是诱导异常凋亡的重要因素。Objective To investigatetherelationshipbetweenabnormalcellularapoptosisof thedecidualtissueduringthe earlystagesof gestationandfailureof pregnancy.Methods Samplesof thedecidualtissuesof earlygestationwereobtained from60healthywomenwithpregnancy,andcellularapoptosiswasassessedby usingterminaltransferase-mediatednick-end labeling(TUNEL).Theexpressionsof FasandFasLinthedecidualtissuesatdifferentstagesof gestationwereexaminedby in situ hybridizationandimmunohistochemistry.Results Onday41of normalpregnancy,evidentapoptosisinthedecidual tissuesoccurredwithsignificantlyenhancedFasandFasLexpressions,whileobviousreductioninboththeapoptoticcellsand FasandFasLexpressionwasobservedon day54.Inwomenwithspontaneousabortion,increasedapoptoticcellswerepresent inthedecidualtissuesaroundday55of gestationwhensignificantlyenhancedFasandFasLmRNAandproteinexpressionsalso tookplace,incomparisonwiththoseof healthypregnantwomenon day54.Conclusion Abnormalapoptosisof thedecidua duringearlygestationmaybeoneof themechanismsforspontaneousabortion,andtheFaspathwaymayplayan importantrole inapoptosisinduction.
