摘 要:目的 探讨次声作用对大鼠视网膜 Müller细胞及血管内皮细胞增殖力的影响。 方法 采用原代培养的大鼠视网膜 Müller细胞和猴视网膜血管内皮细胞 (细胞系 ) ,8Hz、130 d B次声连续辐照 2 h,采用四唑蓝比色实验方法分别于次声作用后 4h、1d、2 d、3d、4d和 5 d6个时间点测定光吸收度值 ,绘制增殖曲线 ,进行统计分析。 结果 8Hz、130 d B次声作用 2 h对原代培养的大鼠视网膜 Müller细胞增殖能力有影响 ,在次声作用后 4h、1d、2 d和 3d4个时间点 ,其光吸收度值与对照组相比差异显著。而猴视网膜血管内皮细胞则对次声不太敏感 ,仅在次声作用后 2 d与对照组的差别有显著性意义。 结论 8Hz、130 d B次声对原代培养的大鼠视网膜 Müller细胞有一定的损伤作用。Objective To investigate the possible effect of infrasound on the proliferation ability of rat's retinal Müller cell and vascular epithelium cell. Methods Primary culture of rat Müller cell and monkey vascular epithelium cell line were inoculated on culture plates and divided into experimental group and control group. The experimental specimens were exposed to infrasound(8 Hz, 130 dB) continuously for 2 h, and then incubated. The OD value of them was measured with MTT assay 4 h, 1 d, 2 d, 3 d, 4 d and 5 d after the exposure. The growth curve was thus obtained, and compared with that of controls. Results For Müller cell culture, the OD value in experimental group was significantly lower than that of control group at 4 h, 1 d, 2 d, 3 d after exposure, while for epithelium cell line, significant difference of OD value between experimentals and controls only found at 2 d after exposure. Conclusion As compared with primary Müller cell, the vascular epithelium cell line is not so sensitive to infrasound. Infrasound has damage effect on the proliferation of rat Müller cell.
