ELISA测定血清LH、FSH的方法学评价

摘　　要：目的　用酶联免疫法测定血中黄体生成素 ( LH)和卵泡刺激素 ( FSH) ,以对该方法的方法学性能作一评价。方法　以临床血清标本作为检测标本 ,用酶联免疫法测定血中的 L H和 FSH,观察其灵敏度、线性试验及线性范围、精密度、回收率 ,并与放免法做比较。结果　 LH、FSH的灵敏度分别为 2 .2 IU/L,1 .3IU/L。线性范围 2 .2～ 2 0 0 IU/L,1 .3～ 1 0 0 IU/L。低、高两种浓度的 LH和 FSH的批内变异分别为8.61 % ,5 .70 %和 9.46% ,6.36% ;批间变异分别为 7.1 4% ,6.0 1 %和 4.46% ,4.69%。回收率 LH为91 .84%～ 1 0 7.66% ,平均 98.1 3% ;FSH为 92 .77%～ 96.88% ,平均 94.5 4 %。与 RIA比较 ,其相关系数LH为 0 .975 9,P<0 .0 0 1 ;FSH为 0 .945 5 ,P<0 .0 0 1。结论　该方法有较高的灵敏度 ,重复性好 ,结果准确可靠等特点 ,与 RIA有良好的相关性 ,且无放射性污染 ,可以替代 RIA用于临床检验。Objective Detection of serum LH and FSH with ELISA and to evaluate this methods.Methods Serum LH and FSH were detected with ELISA.Their of sensitivity.linearity Precision and recovery rate were evaluated and compared with RIA.Results The sensitivity of LH and FSH were 2.2 IU/L and 1.3 IU/L respectively,linear range of LH and FSH were 2.2～200 IU/L and 1.3～100 IU/L.The within run CV for LH and FSH were 8.61%,5.70% and 9.46%,6.36% respectively,between run CV were 7.14%,6.01% and 4.46%,4.69%.The average recovery rate of LH and FSH were 98.13% and 94.54% respectively. Compared 47 samples with RIA,LH and FSH of the relative coefficient( r )0.9759 and 0.9455 respectively. P <0.001.Conclusion The assay has better sensitivity,reproducibility and accuracy.ELISA had a good relativity as RIA.It can act as methods of measuring LH and FSH.
