摘 要:以玉米苹果酸脱氢酶的基因为探针 ,从水稻幼苗cDNA文库筛选得到一条水稻新基因 .它与玉米ZHO2序列的一致性高达 87% ,被命名为RcMDH(ricecytoplasmicmalicdehydrogenasegene) .分析发现RcMDH具有完整的读码框 ,编码 332个氨基酸 ,预计分子质量为 36 .5ku .导入大肠杆菌中表达 。Using the maize MDH gene as probe, a novel rice full length cDNA was cloned through screening a rice seedling cDNA library. It showed 87% identity in nucleic acid sequence to ZHO 2(GenBank No.AF007581)of maize, and therefore was named as rice cytoplasmic malic dehydrogenase gene( RcMDH ). This gene encodes 322 amino acids, with putative molecular weight of 36.5 ku. The open reading frame of RcMDH was subcloned into reconstructed pGEX 4T 1 plasmid which expressed in E.coli with IPTG inducing. The MDH enzyme activity of its product was also certified.
