摘  要:目的研究主动脉平滑肌细胞(aortic smooth muscle cell,ASMC)在三维培养中是否仍能产生弹性蛋白。方法取一周龄SD大鼠胸主动脉,经原代、传代培养后,和胶原、血清及培养液混合形成ASMC胶原凝胶,进行三维培养;培养2周后用Comori醛复红弹性纤维染色和弹性蛋白免疫组织化学染色,检测细胞胶原凝胶中所产生的弹性纤维,并进行电镜观察。结果传代细胞经α肌动蛋白免疫组织化学染色鉴定,98%以上为ASMC。培养2周的ASMC胶原凝胶切片用二种染色方法均可显示有弹性纤维存在,电镜观察细胞超微结构呈典型的合成表型。结论 ASM在三难培养中能够产生弹性蛋白,并形成弹性纤维。Objective To study of aortic smooth muscle cell ( ASMC) can produce elastin in three - dimensional culture. Methods Primary culture of ASMCs were obtained by explants of thoracic aortae from one - week old SD rats. After subcultrue, ASMCs were mixed with collagen, FBS and medium to form three - dimensional gels.After 2 weeks culture, the elastic fiber produced by ASMCs within the gels were examined by Gomori's aldehyde fuchisin elastic fiber staining and elastin immunohistochemistry reaciton, and the cells were observed by electron microscopy. Results Examined by a - Actin immunocytochemistry, more than 98% of the subcultred cells were ASMCs. After 2 weeks culture, elastic fiber were detected in the ASMCs - collagen gels by both methods.The cells presented typical synthetic ultrastiuctures. Conclusion ASMC in three - dimentional culture can synthesize elastin and produce elastic fiber.
