甲型肝炎病毒吕8株基因型的研究

摘　　要：目的　对甲型肝炎病毒吕 8株第 2 5代细胞适应株的部分基因序列作基因克隆分析。方法　采用逆转录聚合酶链反应法从细胞培养产物中克隆病毒并进行核苷酸序列分析。结果　吕 8株与HM175 /7MK 5的核苷酸 /氨基酸同源性最高为 98% /96 % ,基因型属于ⅠB亚型。结论　吕 8株与中国地区以前报道的大多数甲肝病毒株有较大差异 。Objective To analyze the genotypes of a hepatitis A virus Lü8 strain passaged 25 times in cells. Methods Partial genome of a hepatitis A virus, Lü8 strain, was amplified with RT PCR. The PCR products were cloned into pGEM T Easy vector and sequenced. Results The results showed that the most extensive nucleotide/amino acid homology was identified between Lü8 and HM175/7MK 5 strain with 98%/96% and phylogenetic analysis indicated that Lü8 strain of the hepatitis A virus belonged to genotypesⅠB. Conclusion Lü8 isolated shows much difference from most other HAV strains which had been reported in China. [
