摘  要:目的 探讨一种简便、有效的红细胞长期保存的方法。方法 红细胞在 4 0℃、 80℃及 196℃ (液氮 )不同温度下长期保存 18个月后 ,分别测定冻存前后红细胞平均体积 (MCV)、红细胞渗透脆性 (RFT)及红细胞免疫功能 (RBC C3bRR、ERPN)。结果 红细胞 4 0℃长期冻存后 ,MCV和RFT较冻存前显著增高 (P <0 0 5 ) ,RBC C3bRR和ERPN较冻存前显著降低 (P <0 0 5 )。红细胞 80℃、 196℃长期冻存后 ,MCV、RFT、RBC C3bRR及ERPN较冻存前无明显改变 (P >0 0 5 )。结论 红细胞经 80℃、 196℃长期冻存后 ,其形态、结构和免疫功能保存良好。 80℃低温冻存是一种简便。Objective To research a simple and effective method of erythrocytes cold storage for a long time.Methods Red blood cells were cryopreserved in 40℃、 80℃ and 196℃(liquid nitrogen)for 18 months, before and after cold storage, mean corpuscular volume (MCV)、red blood cells fragility test(RFT)、red blood cells C 3b receptor rosette rate(RBC C 3b RR) and enhancing rosette rate of phagocytosis of red blood cells on neutrophils (ERPN)were measured.Results After 40℃ cryopreservation for 18 months, MCV and RFT of red blood cells were significantly increased( P <0 05),RBC C 3b RR and ERPN of erythorcytes immune functions were significantly decreased( P <0 05)compared with before cold storage. After 80℃ and 196℃ long cryopreservation, there was no great difference in erythrocyte MCV、RFT、RBC C 3b RR and ERPN( P >0 05) compared with control groups.Conclusion It was suggested that, after 80℃ and 196℃ long cryoperservation, there was no marked changes in shape、structure and immune functions of red blood cells. 80℃ cryopreservation is a simple and effective methods of erythrocytes cold storage for a long time.
