摘 要:本文以锇酸—二甲基亚砜—锇酸冷冻割断法(ODO法)制备菊科和木兰科几种植物花粉样品,在扫描电镜下,可从整朵花、花药和花粉的割断面,观察花药的位置及断面形状、花粉在药室中的分布和着生状况、花粉壁的层次和结构、萌发孔结构及花粉粒内外之间的联系、绒毡层形态及花粉发育、花粉内细胞核、核膜、线粒体、内质网等的亚显微三维结构,形态真实完整,图象清晰,说明ODO制样方法将扫描电镜能获得三维空间立体图象及放大范围广的特点,成功地应用于花、雄蕊内部结构及花粉粒细胞器亚显微结构的观察,从而在方法学上,为孢粉学研究提供了新的有效手段。The specimen of pollen grains of several species on Compositae andMagnoliaceae were prepared by ODO freeze cracking method. Under SEM,the various arrangement of pollen grains within locules, the layers and structure of aperture and pollen wall, the inside of pollen connectedwith the outside have been observed from the cracked surface of flower,anthers and pollen grains. Not only the morphological characters of flo-wer, anther and pollen but also the three dimensions of substructure ofnucleus, nuclear membrane, and organella such as mitochondria, endo-plasmic reticulum in pollen were shown. The live, clear images of spe-cimen are complete success, thus provided an effective approach to stu-dy the morphology and structure of pollen.
