摘  要:目的 建立利钾尿肽 (KP)放射免疫分析法 (RIA)并用于临床。方法 将KP和甲状腺球蛋白连接制成免疫原 ,免疫新西兰白兔获得高特异性KP抗体 ,采用Iodogen法制备12 5I KP ,经高效液相色谱纯化后其比活度为 6 4.75TBq/mmol。测定尿毒症、高血压、急性心肌梗死患者及正常对照者的血浆KP含量。结果 方法的测定范围是 5 0~ 30 0 0pmol/L ,IC50 =92 5pmol/L。对照组血浆KP含量为 (2 6 0 82± 2 9 45 )pmol/L ,而尿毒症、高血压、急性心肌梗死患者的KP含量均显著升高。结论 KPRIA的建立为开展相关研究提供了一种灵敏。Objective To develop a sensitive and specific radioimmunoassay of kaliuretic peptide (KP) and apply it in clinical research. Methods KP and thyreoprotein were conjugated and the conjugate was injected into New Zealand white rabbits. A high sensitive and specific antibody of KP was obtained. 125I-KP was prepared by Iodogen method and was purified by HPLC. Its specific activity was about 64.75 TBq/mmol. The plasma concentrations of 125I-KP of the healthy volunteers and the hypertensive, uraemia and acute myocardial infarction patients were determined. Results The assaying range was 50~3 000 pmol/L, IC 50=925 pmol/L. The plasma KP value of the healthy volunteers was (260.82±29.45) pmol/L (n=25) and the KP levels in the hypertensive, uraemia and acute myocardial infarction patients were significantly higher than that in normal subjects. Conclusion This developed assay could be a usable technique in related research of KP.
