摘  要:目的探讨大蒜对松花江水有机提取物致癌活性的阻断作用及其机制,为进一步开发和利用大蒜预防肿瘤提供依据。方法采用体外试验 ,观察大蒜浸液对松花江水有机提取物诱导的BALB/C胚胎成纤维细胞恶性转化及其DNA损伤的影响 ,并通过检测大蒜水浸液对转化细胞p53、p16、p21表达和细胞凋亡的作用 ,探讨其阻断作用的机理。结果大蒜水浸液可通过诱导细胞中p53、p16和p21的表达 ,进而引起受损伤细胞的凋亡而抑制江水有机提取物诱发的转化。对已经发生恶性变的细胞 ,大蒜水浸液没有诱发细胞凋亡的作用 ,3种蛋白的表达率也未见升高。结论大蒜水浸液对松花江水有机污染物的致癌活性具有阻断作用。Objective To explore the blocking effects of garlic on carcinogenic activity of the organic extracts from water in Songhua River and its mechanism, and to provide the scientific evidence for further exploration and utilizing the resource of garlic to prevent the carcinoma. Methods Using the method of BALB/C embryonic fibroblast cultivation in vitro, the influence of water logging extracts from garlic on malignant cell transformation and DNA injury induced by organic extracts of water in Songhua River were observed. The mechanism of blocking effects was studied by detecting the effects of water logging extracts from garlic on the expression of p53, p16 and p21 protein in transformed cells and the cell apotosis. Results The cell transformation induced by organic extracts from water of Songhua River was inhibited by water logging extracts from garlic, which could induce the expression of p53, p16, and p21 protein and apoptosis in the cell with DNA injury. No occurence of cell apoptosis and no significant increases of expression of 3 kinds of protein induced by water logging extracts from garlic were found in malignant transformed cells. Conclusion It indicated that water logging extracts from garlic could block the carcinogenic activity of organic extracts from water of Songhua River.
