摘  要:目的探讨枸杞提取物拮抗六价铬的致突变作用。方法采用人外周血淋巴细胞程序外DNA合成 (UDS)试验检测了0.02、0.01和0.005g/ml枸杞提取物对重铬酸钾 (含Cr6 +0.31μg/ml)所致淋巴细胞程序外DNA合成的影响 ,同时检测枸杞提取物与重铬酸钾的加入顺序对UDS的影响。结果阳性对照 (重铬酸钾 ,含Cr6 +0.31μg/ml)组CPM值显著高于阴性对照 (蒸馏水 ) ,P<0.01。枸杞提取物与重铬酸钾同时加入以及二者间隔2h先后加入均显示枸杞提取物浓度与CPM值呈显著负相关 (P<0.05) ,在上述不同加入方法中 ,枸杞提取物的各浓度均显示CPM值显著低于阳性对照。结论枸杞提取物具有拮抗Cr6 +的致突变作用。Objective To study the antagonistic effect of organic extracts of Fruit of Chinese Wolfberry (FCW) on chromium (Ⅵ) _induced mutation. Methods Using unscheduled DNA synthesis (UDS) assay, the effects of 0.02 g/ml, 0.01 g/ml and 0.005 g/ml organic extracts of FCW on UDS induced by potassium bichromate [0.31 μg/ml Chromuim (Ⅵ)] in human peripheral blood lymphocytes were determined. The effects of the order of adding organic extract of FCW and potassium bichromate into cultured lymphocyte suspension on UDS were observed at the same time. Results The value of CPM revealed a significantly higher level in positive control group exposed to 0.31 μg/ml chromium (Ⅵ) compared with that in negative control group exposed to disstilled water (P<0.01). In the comparative experiments with different methods for adding the substances to be tested: simultaneously adding the organic extracts of FCW and potassium chromate into the cultured lymphocyte suspension, and adding the organic extracts of FCW first and potassium chromate afterward or in turn at intervals of 2 hours, the values of CPM revealed a significant negative correlation with the exposure levels of organic extracts of FCW (P<0.01), and each value of CPM in all FCW groups was significantly lower than that of positive control group. Conclusion FCW revealed significant antimutagenic effect on chromium (Ⅵ).
