摘 要:目的:研究益肝浓缩煎剂、强肝胶囊、丹参复方(丹参+当归+水蛭)、当归、水蛭对大鼠肝纤维化降解作用,并对其机制进行初步探讨。 方法:将120只(?)SD大鼠随机分成8组,即正常对照组(A组)、模型组(B组)、模型自然恢复组(C组)、中药治疗组(D-C组)。采用400mL·L^(-1)四氯化碳(CCl_4)_(sc)诱导大鼠肝纤维化模型,造模结束后分别给予相应药物进行治疗,持续10wk。测定肝组织羟脯氨酸(Hyp)、丙二醛(MDA)含量,光镜观察肝脏病理形态变化。 结果:益肝浓缩煎剂、强肝胶囊及丹参复方治疗组Hyp、MDA显著降低(Hyp 124±20μg·g^(-1)肝粉,130±18μg·g^(-1)肝粉,149±16μg·g^(-1)肝粉 vs 190±29μg·g^(-1)肝粉P<0.05;MDA13.8±1.6umol·L^(-1),14.1±2.5 umol·L^(-1),15.4±1.5 umol·L^(-1) us 23.1±2.4 umol·L^(_1)P<0.05),胶原沉积明显减轻(P<0.05);当归治疗组Hyp、MDA显著下降(Hyp 163±16μg·g^(-1)肝粉vs 190±29μg·g^(-1)肝粉P<0.05,MDA18.7±2.4umol·L^(-1) vs 23.1±2.4umol·L^(-1)P<0.05),胶原沉积无减轻(P>0.05);水蛭治疗组各项指标均无好转(Hyp179:25μg·g^(-1)肝粉 vs 190±29μg·g^(-1)肝粉P>0.05,MDA21.9±2.6umol·L^(-1)。vs 3.1±2.4umol·L^(-1)P>0.05)。 结论:益肝浓缩煎剂、强肝胶囊?AIM: To study the degradation of concentrated Yigan decoction( Yigan), Qianggan capsule (Qianggan), formula of Radix Salviae Miltiorrhizae ( i. e. Radix Salviae Miltlorrhlzae + Radix Angelicae Sinensis + Hirudo ) (formula of RSM), Radix Angelicae Sinensis (RAS) and Hirudo on hepatic fibrosis in rats as well as their relative mechanisms.METHODS:Total 120 male SD rats were randomly divided into eight groups, i. e., normal control group (group A), pathological model group (group B) , model of natural recovering group (group C), treatment groups(group D-H). 400 mL · L-1 carbon tetrachloride (CCl4) oily solution was injected subcutaneously to rats twice a week for a period of 8 weeks. The relative herbs were given after model establishment in each treatment group and the administration lasted for 10 weeks. Liver hydroxyproline (Hyp) and malondialdehyde (MDA) level were determined and histopathological changes were observed by optical microscope.RESULTS: In Yigan, Qianggan and formula of RSM groups, the contents of Hyp and MDA (Hyp 124 ± 20 μg · g-1 liverpowder, 130 ± 18μg· g-1 liver powder, 149 ± 16μg· g-1 liver powder vs 190±29μg·g liver powder;MDA13.8± 1.6 umol· L-1 ,14.1 ± 2.5 umol · L-1, 15.4 ± 1.5 umol - L-1 vs 23.1 ± 2. 4 umol · L-1 ) as well as degrees of hepatic fibrosis were markedly ( P < 0.05) reduced respectively, compared with those of group C. Hyp and MDA were markedly reduced in RAS group (Hyp 163 ± 16μg·g-1 liver powder vs 190 ± 29 μg· g-1 liver powder P < 0.05, MDA18. 7 ± 2.4 umol · L-1 vs 23.1 ± 2.4 umol · L-1 P < 0.05) while no statistical change was noted in degree of hepatic fibrosis ( P > 0. 05). In Hirudo group there was no significant decrease in all respects (Hyp179 ± 25μg ·g-1liver powder vs 190 ± 29μg· g-1liver powder P> 0.05,MDA 21.9 ±2.6 umol·L-1 vs23.1±2.4umol·L-1 P>0.05).CONCLUSION: Yigan, Qianggan, and formula of RSM can resolve the collagens accumulated in liver of rats, superior to RAS, and hepatic fibrosis at early stage can be reversible in th
