复方仙灵脾注射液对大鼠局灶性脑缺血血流变和自由基的影响

摘　　要：目的 :观察具有补肾益气、活血通络功效的中药复方仙灵脾注射液对脑缺血性大鼠模型的体重、神经症状、被动性条件反射、血液流变性、血小板聚集、自由基代谢、脑梗塞面积的指标影响。方法 :采用结扎大鼠右侧大脑中动脉 (RightMiddleCerebralArteryOcclusion ,R -MCAO )造成局灶性脑缺血模型。将SD大鼠 60只 (雌雄各半 ,6月龄 )随机分为 6组 ,每组 10只 ,即假手术组、模型组、川芎嗪对照组、复方仙灵脾大、中、小剂量组。结果 :复方仙灵脾注射液能明显改善MCAO大鼠神经学症状 ,显著延长被动性条件反射潜伏期 ,减少错误次数 ,降低全血粘度 ,降低血小板聚集率 ,使血浆SOD升高 ,MDA下降 ,缩小脑梗塞面积等。结论 :复方仙灵脾注射液在改善血液流变性 ,增加脑血流量的同时 ,又能清除自由基 ,改善脑功能 ,减轻急性脑梗塞时缺血损伤。Objective:With rats as experiment object,this study is purposely to sbserve the effects of FuFang XianLingPi(FFXLP) injection ,a combination of traditional Chinese medical herbs which has a fuction of tonifying the kidney and invigorationing qi and promoting blood circulation to remove obstruction in the channels,on focal cerebral ischemia was produced in rats by the occlusion of right middle cerebral artery,the gherapeutic action of FFXLP injection on focal cerebral ischemia was studied by investigating the change of weight ,neurologic symptoms,passive conditional reflex,hemorheology,platelet aggregation rate,metabolism of free-radical and infarct size of brain.Methods:The focal cerebral ischemia rat model was made by the occlusion of right middle cerebral artery.60 Sprague-Dawsley(SD)rats(30) males and 30 females,6 month old)were randomly divided into six groups;Pseudo-operation groups(n=10),model groups(n=10),tetramethylpyrazine injection controls group(n=10),among treatment group of FFXLP injection large dose group(n=10),middle dose group(n=10)and small dose group(n=10).Results:the FFXLP injection conld not only improve obviously neurologic symptoms,prolong latent period of the passive conditional reflex,lower blood viscosity,lower platelet aggregation rate,increasing SOD activity,but also in decteasing the content of MDA and lessen infarct size of brain.Conclusion:FFXLP injection can improve the blood rheology and increase the olume of cerebral blood flow at the smme time it is able to eliminate oxygen free gadicals,improve cerebral functions,and relieve ischemia damage in acutely cerebral infariction.
