摘 要:目的 探讨甲基汞对小鼠胸腺淋巴细胞DNA损伤的作用。方法 采用单细胞凝胶电泳技术进行检测。结果 1 2 0 0~ 1 2LD50 ,剂量组的甲基汞均可引起淋巴细胞DNA不同程度电泳迁移 ;A组 ( 1 2 0 0LD50 )、B组 ( 1 2 0LD50 )、C组 ( 1 2LD50 )DNA平均迁移长度与对照组之间差异有显著性 (P <0 0 5 ) ,并且C组与A、B两组之间差异也有显著性 (P <0 0 0 1) ;B组尾直径 头直径、尾面积 总面积之比均与A组差异有显著性 (P <0 0 5 )、C组各项之比均与A、B组差异有显著性 (P <0 0 5 )。结论 甲基汞引起DNA链断裂损伤 ,随着甲基汞剂量的增加 ,对淋巴细胞DNA损伤加重。Objective To study the effects of different doses of MeHg on DNA damage of thymocytes in mice.Methods Using the technique of single cell gelelectrophoresis. Results There were different degree DNA migrations of lymphocytes exposed to 1/200LD 50 doses of the MeHg; The length of DNA migrations of lymphocytes exposed to groupA(1/200LD 50 ),groupB(1/20LD 50 ),groupC(1/2LD 50 )were significantly different to the control group(P<0.05),and the difference was statistically significant between groupC and the other groups(P<0.001)the rate of tail diameter and head diameter of group B was significantly different to group A(P<0.05);and the rate of tail areas and total areas of group B was significat different too (P<0.05);all the rate of group C was significant to the other groups(P<0.05).Conclusion MeHg caused single-stranded DNA break,and with the dose of MeHg increasing,the degree of DNA damage hardened.
