摘 要:目的 :比较黄芪和非创伤性肢体缺血两种预处理方法对大鼠离体心肌再灌注损伤的保护作用。方法 :采用雄性大鼠 ,在Langendorff装置上对离体心脏进行灌流。预灌 15min后 ,缺血 15min ,随后 15min再灌注。观察先用含黄芪注射液 ( 10mg/L)预灌在相应点分别测定冠脉流出液和心肌匀浆中SOD的活性和MDA含量 ,同时记录心肌细胞的单相动作电位 (MAP)和心肌收缩张力曲线。结果 :黄芪预处理和非创伤性肢体缺血预处理能显著降低再灌注心律失常发生率 ,显著降低心肌组织中MDA含量 ,提高SOD活性 ,稳定心肌细胞的膜电位和肌张力。结论 :黄芪预处理对大鼠离体心肌再灌注损伤有明显的保护作用 ,可能是通过增强心肌的抗氧化能力、稳定心肌的膜相结构等途径而实现的。AIM: To study the protective effects of Astragalus and non wounded ischemic preconditioning on ischemic/reperfusion injury in isolated perfused rat hearts. Methods: 32 male SD rats, weighting 250±30g, were randomly divided into 4 groups: normal control group (NC, n=8), ischemic/preconditioning group (I/R, n=8), Astragalus preconditioning groups (APC, n=8) and non wounded legs ischemic preconditioning group (N WIPC, n=8). The activities of superoxide dismutase (SOD) and Ca 2+ Mg 2+ ATPase, malondialdehyde (MDA) contents of efflux from cononary vessel and myocardium, myocardium monophasic action potential and contractile force were measured before ischemia, 15min after ischemia and 5, 15min after reperfusion. Results: Compared with I/R group, Astragalus preconditioning and non wounded legs ischemic preconditioning could reduce the incidence of reperfusion arrhythmias (P<0.05), decrease the content of MDA of myocardium (P<0.01), enhance the activities of SOD (P<0.05), stabilize myocardial membranous potential and contractile funtion. Conclusions: It indicates that Astragalus and non wounded legs ischemic preconditioning have some definitely capabilities on anti reperfusion injury in isolated perfused rat hearts. It may be related to the strength of antioxidation, the stability of myocardial membranous structure.
