摘 要:目的 探讨血管紧张素转换酶抑制剂 (ACEI)苯那普利对糖尿病大鼠肾脏细胞凋亡及凋亡相关基因表达的影响。方法 单侧肾切除大鼠ipSTZ诱发糖尿病模型 ,每日ig苯那普利 ( 10mg·kg- 1 ) ,共 12周。采用原位末端标记法检测肾脏细胞凋亡情况 ,流式细胞术和免疫组化检测肾皮质凋亡相关基因Fas和Fas L的表达。结果 糖尿病组较对照组肾小球、肾小管凋亡细胞数明显增多 ,Fas和Fas L的表达亦显著增强 ;苯那普利治疗组较糖尿病组凋亡细胞数减少 ,Fas和Fas L的表达减弱。结论 苯那普利可能通过影响凋亡相关基因Fas和Fas L的表达而抑制肾脏细胞凋亡 。AIM To investigate the effects of angiotensin converting enzyme inhibitor (ACEI) on apoptosis and the expression of Fas and Fas L in the kidney of diabetic rats. METHODS Uninephrectomized Spraque Dawley rats were used to induce diabetes by intraperitoneal injection of streptozotocin (65 mg·kg -1 ). Benazepril (10 mg·kg -1 ) was given daily by gavage from the next day of the induction to diabetes for 12 weeks. Apoptosis was evaluated by means of terminal deoxynucleotidyl transferase mediated d UTP nick end labeling (TUNEL). Flow cytometry and immunohistochemistry were used to detect the expression of Fas and Fas L. RESULTS Compared with those in the kidneys of control group, apoptotic cells were more in number and the expression of Fas and Fas L was higher in the diabetic kidneys ( P <0 05). The number of apoptotic cells and the level of expression of Fas and Fas L were reduced by benazepril treatment ( P <0 05). CONCLUSION Angiotensin converting enzyme inhibitor benazepril showed some renal protective effect on diabetic nephropathy, partly through inhibiting apoptosis by down regulating Fas and Fas L expression.
