胰岛素对自发型高血压大鼠血管平滑肌细胞TGFβ及其受体的调节作用(英文)

摘  要:目的 :从细胞增生、转化生长因子 β1和受体表达等方面 ,探讨胰岛素对自发型高血压大鼠血管平滑肌细胞(SHRVSMC)和正常血压Wista -Kyoto大鼠血管平滑肌细胞 (WKYVSMC)的影响异同。方法 :采用组织移植法培养SHR和WKY大鼠胸主动脉VSMC ;用细胞计数仪和流式细胞仪分别检测细胞增生情况 ;TGFβ及其受体的mRNA水平利用定量RT -PCR技术进行检测。结果 :(1)胰岛素加强SHRVSMC的增生 ,而不影响WKYVSMC的增生。胰岛素加强SHRVSMC的增生 ,且呈剂量依赖方式 ;(2 )以 2 0 %小牛血清培养基培养VSMC ,SHRVSMC的S期百分率为 31 4 4 % ,而WKYVSMC的S期百分率仅为 19 86 % ;以 2 %小牛血清培养基培养VSMC ,SHRVSMC的G0 /G1和S期百分率分别为 73 2 3%和 9 35 % ,加入胰岛素后 ,SHRVSMC的G0 /G1降低为 6 7 5 8% ,S期百分率则增加到 15 6 4 %。但是 ,加入胰岛素前后 ,WKYVSMC各期百分率无明显变化 ;(3)RT -PCR分析结果表明 ,生长静止的SHRVSMC和WKYVSMC均有TGFβ1、Ⅰ型和Ⅱ型TGFβ受体的表达 ,但SHRVSMC的TGFβ1、Ⅰ型TGFβ受体的表达量高于WKYVSMC的TGFβ1、Ⅰ型TGFβ受体的表达量 ,胰岛素加强了WKYVSMC的TGFβ1、Ⅰ型TGFβ受体的表达 (P <0 0 1和P <0 0 5 ) ,而削弱了SHRVSMC相应分子的表达 (P <0 0 1和P <0 0 5 ) ,AIM: To explore the effect of insulin on the proliferation and expression of transforming growth factor β 1 (TGF β 1) and its receptors in aortic vascular smooth muscle cells (VSMC) from spontaneously hypertensive rats (SHR) and from the normotensive Wistar-Kyoto(WKY) rats. METHODS: VSMCs from aortas of WKY rats and SHR were prepared by an explant method. The effect of insulin on VSMC proliferation were measured by cell counting and flow cytometry analysis, respectively. Quantitative reverse transcription polymerase chain reaction (RT-PCR) technique was used to probe TGF β 1 and its receptor mRNA on VSMC. RESULTS: Insulin enhanced the proliferation of VSMC from SHR in a dose-dependent manner ( P< 0 01), with no effect on VSMC from WKY rats. The proportion of cells in S phase were increased from 9 35% to 15 64% by insulin in VSMC from SHR but not in VSMC from WKY. RT-PCR analysis revealed that the expression of TGF β 1 and type I TGF β receptor were enhanced by insulin in VSMC from WKY rats ( P< 0 01 and P< 0 05, respectively), but both decreased in SHR derived cells ( P< 0 01) The expression of type II TGF β receptor mRNA were not affected by insulin in VSMC of either strain ( P> 0 05). CONCLUSIONS: The responses to insulin of TGF β 1 and its receptor in SHR and WKY are significantly different. This abnormal regulation by insulin may be associated with an exaggerated proliferative response in SHR derived cells.
