摘  要:目的 探讨TGF β对胎兔颅骨成骨样细胞体外成骨作用及BMP蛋白表达的影响。 方法 采用ALP活性测定及骨结节计数方法观察TGF β对胎兔颅骨成骨样细胞体外的成骨作用 ,BMP McAb免疫组化染色及其定量分析观察TGF β对胎兔颅骨成骨样细胞BMP蛋白表达的影响。结果 TGF β( 3~ 7d)对ALP活性有促进作用 ,并呈一定的时效和量效关系。TGF β对骨结节形成低剂量时有促进作用 ,高剂量时有较强的抑制作用 ;TGF β对成骨细胞BMP蛋白表达表现为抑制作用 ;二者均有显著性差异。 结论 TGF β对胎兔颅骨成骨样细胞体外成骨及BMP蛋白表达表现为双相作用。Objective To explore the effects of TGF-βon the osteogenosis and the expression of bone morphogenetic protein (BMP) of primary osteoblast-like cells derived from fetal rabbit calvarium in vitro . Methods The effects of TGF-β on the osteogenosis of primary osteoblast-like cells were investigated with alkaline phosphatase activity assay and bone nodule counting. The effects of TGF-β on BMP expression were tested with immunohistochemical staining and quantitative analysis. Results TGF-β given for 3-7 d increased ALP activity in a dose- and time-dependent manner. It was found TGF-β enhanced the bone nodule formation at low concentration, but inhibited it at high concentration. TGF-βinhibited the BMP protein expression, which increased along with the dose. Conclusion TGF-β showed bi-directional effects on the osteogenesis and BMP protein expression of rabbit osteoblast-like cells derived from fetal calvarium in vitro .
