摘  要:目的 研究CagA阳性Hp培养滤液对人胃粘膜上皮细胞 (GES 1)作用的机制。方法 将CagA阳性Hp培养滤液诱导恶性转化的GES 1细胞与CagA阴性Hp培养滤液处理的GES 1细胞进行mRNA差异显示 ,以寻找Hp致胃癌发病的相关基因。结果mRNA差异显示片断之一命名为PFN1,仅在Hp(CagA+ )培养滤液处理的GES 1细胞中表达 ,斑点杂交结果与之一致。与GeneBank资料序列同源性比较分析提示PFN1与泛素活化酶E1同源性 99% ,即为人类泛素活化酶E1基因的一部分。结论 泛素 蛋白水解酶复合体通路中的泛素活化酶E1基因可能参与了Hp的CagA诱导的胃上皮细胞恶性转化过程。Objective To study the mechanism of malignant transformation induced by cagA positive broth culture filtrates of Helicobacter pylori (Hp) on human gastric epithelial cell line GES-1. Methods CagA related-gene was searched with mRNA differential display technique by using GES-1 cells after they were treated with CagA positive and negative broth culture filtrates of Hp. Results The result of mRNA differential display showed that there were significant differences in gene expressions between GES-1 cells treated with CagA positive and negative filtrates of Hp. One fragment, PFN 1 as named, was only expressed in the GES-1 cells with the treatment of CagA positive Hp filtrates. Dot blotting analysis showed similar results. With aid of Genbank homology analysis program, the sequence of PFN1 had 99% homology with the homo sapiens ubiquitin-activating enzyme E1 gene. Conclusion Human ubiquitin-activating enzyme E1, a ligase in ubiquitin-proteasomes pathway, may be involved in the malignant transformation of gastric epithelial cells induced by CagA gene of Hp.
