摘  要:探讨核因子κB(NF κB)对哮喘患者T淋巴细胞HO 1表达的转录调节机制。分离 18例急性发作期哮喘患者外周血T淋巴细胞 ,并分成 3组培养 :对照组、加入NF κB激动剂肿瘤坏死因子 α(TNF α)组、同时加入TNF α和NF κB抑制剂二硫代氨基甲醇吡咯烷 (PDTC)组。培养 6h后留取细胞 ,用逆转录聚合酶链反应 (RT PCR)检测血红素氧合酶 1(HO 1)的mRNA。培养 2 4h后留取细胞用Western印迹法检测HO 1的表达。发现TNF α组T淋巴细胞HO 1蛋白和mRNA表达水平显著高于对照组 (q =4 4 4 8、2 9 94 ,P均 <0 0 1) ,而同时加入TNF α和PDTC培养组T淋巴细胞HO 1蛋白和mRNA表达水平显著低于TNF α组 (q =4 3 2 3、2 7 99,P均 <0 0 1)。可见哮喘患者T淋巴细胞HO 1基因转录可能是通过激活NF κB进行调控。T淋巴细胞NF κBTo explore the regulative effect of nuclear factor kappa B (NF κB) on the expression of heme oxygenase 1 (HO 1) in T lymphocytes. 16 asthmatic patients participated the study, T lymphocytes were isolated and purified from blood and divided into 3 groups, cells of 1st group were control group, cells of 2nd group were stimulated with Tumor necrosis factor alpha(TNF α) and cells of 3rd group were stimulated with TNF α and pyrrolidine dithiocarbamate(PDTC). HO 1 protein in T lymphocyte was measured by Western blot,the expression of HO 1 mRNA was examined by reverse transcription polymerase chain reaction (RT PCR). The results showed that HO 1 protein, HO 1 mRNA of asthmatic T lymphocytes stimulated with TNF α were significantly higher than those of asthmatic T lymphocyte stimulated with TNF α and PDTC (q=43.23?27.99, P <0.01, respectively), and were also significantly higher than that of control group (q=44.48?29.94, P <0.01, respectively). These results suggested that asthmatic T lymphocytes increased the expression of HO 1 that was regulated by activating NF κB. The oxidizing activation of NF κB HO 1 signal pathway of T lymphocytes may be one of important mechanisms of asthma.
