摘 要:目的探讨急性放射性皮肤溃疡伤口收缩迟缓的机制。方法采用雌性Wistar大鼠,以50Gy60Coγ射线单次局部照射建立急性放射性皮肤溃疡动物模型,以手术方法建立单纯皮肤伤口模型,观察伤口纤维组织的改变及α-平滑肌肌动蛋白的表达,并检测伤口组织内羟脯氨酸含量。结果照射后3~11d胶原纤维出现肿胀、融解、断裂,排列紊乱,照后14~55d溃疡内胶原纤维较单纯伤口内减少,网状纤维渐增多。照射后14~55d放射性皮肤溃疡内α-平滑肌肌动蛋白的表达较单纯伤口内减弱,羟脯氨酸含量减少。结论急性放射性溃疡组织内胶原合成减少、纤维组织结构异常以及肌成纤维细胞数量减少与急性放射性皮肤溃疡收缩迟缓的机制有关。Objective To study the mechanism of wound contr action retardation of acute radiation-induced skin ulcers.Methods Fe-male Wistar rats were used to set up an animal model of simple skin wounds,a nd another model which was locally ir radiated with 50Gy 60 Co γ-rays was used.Immunohistochemical staining,fibrous staining,spectrophotometer were used to detect the changes inа-smooth muscle actin,collagen fibers and hydroxyp roline in these models.Results The collagen fibers showed swelling,thawing,fragmentation and de-rangement during 3to 11days after ir radiation.Collagen fibers,hydroxyproline and the expression ofа-smooth muscle actin were decreased compared to the simple wounds,while reticular fibers were increased and elastic fibers were hardly seen in th e radiation ulcers during 14to 55days after radiation.Conclusion The disarrangement of fibrous structure,reduction of collagen synthesis and the decreased number of my-ofibroblasts were relevant to the wo unds contraction retardation of acu te radiation-induced skin ulcers.
