急性坏死性胰腺炎大鼠核因子-κB的表达及其意义

摘　　要：目的 :观察 NFκB在大鼠急性坏死性胰腺炎 (ANP)时的动态表达 ,探讨核因子 κΒ(NFκB)在胰腺炎炎症反应中的信号传导过程。方法 :12 6只 SD大鼠随机分为正常对照组、ANP组和地塞米松治疗组 ,利用改良的 Pfetter法复制 SD大鼠 ANP模型 ,采用免疫组织化学的方法 ,动态观察 3组大鼠胰腺组织中 NFκB的表达 ,并观察胰腺病理变化。结果 :病理观察显示 ,ANP组大鼠胰腺组织有明显坏死、出血及炎症细胞聚集 ,经地塞米松治疗后炎症反应减轻。胰腺组织中 NFκB在 ANP后 1小时开始表达 ,3小时达高水平 ,6小时后下降 ;而正常对照组无明显表达 (P<0 .0 1)。地塞米松治疗组 NFκB也有表达 ,但弱于 ANP组 (P<0 .0 1)。结论 :信号转录因子 NFκB参与了大鼠 ANP的炎症反应 ,地塞米松对Objective:To investigate the change in nuclear factorκB(NFκB) expression and its potential role in infammation signal transduction in rats following acute necrotizing pancreatitis(ANP).Methods:The modified Pfetter′s method was used to reproduce ANP model in rats.One hundred and twentysix SD rats were randomly divided into normal controls,ANP group and dexamethasone treatment group.The expression of NFκΒ in pancreatic tissue was determined by immunohistochemical staining.Results:The histological examination revealed obvious necrosis,hemorrhage,as well as inflammatory cell infiltration in pancreatic tissue after ANP.In the ANP group,NFκB in pancreatic tissue was expressed at 1 hour,peaking at 3 hours and decreasing at 6 hours,while there was no marked expression in control group( P <0 01).Treatment with dexamethasone could significantly inhibit the expression of NFκB compared to the ANP group( P <0 01). Conclusions: NFκB is involved in the inflammation response of ANP and treatment with dexamethasone could inhibit the expression of NFκB.
