急性胰腺炎大鼠胰腺细胞凋亡与胰腺炎症细胞的关系

摘　　要：目的 :探讨急性胰腺炎 (AP)胰腺细胞凋亡的可能机制。方法 :胆胰管逆行加压注射 4 %的牛磺胆酸钠诱导大鼠AP ,建立模型 ,应用末端脱氧核苷酸转换酶 (TdT)介导的原位末端标记 (TUNEL)法检测术后 3h、6h和 12h胰腺细胞凋亡情况 ,并对胰腺炎症细胞进行分类记数。结果 :胰腺炎组术后 6h胰腺凋亡细胞显著高于 3h和 12h(3hP <0 .0 5 ,12hP <0 .0 1) ;白细胞进行性增多 ,6h巨噬细胞百分比明显高于 3h和 12h(3hP <0 .0 5 ,12hP <0 .0 1) ,中性粒细胞百分比显著低于 3h和 12h(3hP <0 .0 5 ,12hP <0 .0 1)。结论 :AP胰腺细胞凋亡可能与炎症细胞的不同种类有关。Objective:To investigate the mechanisms of pancreatic acina cell apoptosis in rats with acute pancreatitis(AP).Methods:The model of AP was induced by retrograde injection of 4% sodium taurocholate into biliopancreatic duct in rats. A terminal deoxynucleotidyl transferase mediated dUTP biotion nick end labeling(TUNEL) was used to detect apoptosis. Leukocytes were identified by morphological features on paraffin sections obtained from the pancreas stained with HE.Results:Apoptosis index of AP rats at 6 h was higher significantly than that at 3,12 h(3 h P<0.05,12 h P<0.01,respectively). The types of leukocyte differed significantly in AP rats. Compared with that at 3,12 h,the percentage of macrophages at 6 h were significantly higher(3 h P<0.05,12 h P<0.01,respectively),while the percentage of neutrophils at 6 h were significantly lower(3 h P<0.05,12 h P<0.01,respectively).Conclusion:Pancreatic acina cells undergoing apoptosis may result from differences in inflammatory cell infiltration in AP.
