河蟹颤抖病病毒的分离纯化及其动物致病性试验

摘　　要：将人工复制发病的河蟹 (中华绒螯蟹 )组织匀浆 ,离心上清经聚乙二醇 (PEG 6 0 0 0 )沉淀 ,再经分子筛层析 ,分部收集分别测定 2 5 6～ 30 0nm的吸光值。各收集峰用PEG2 0 0 0 0 ,于 4℃浓缩 ,制作负染标本经透射电镜观察 ,发现在第一峰浓缩液中存在大量球状病毒颗粒 ,大小 5 0～ 10 0nm。用纯化的病毒进行动物致病性试验 ,接种石蟹 (锯齿华溪蟹 )发生“颤抖病” ,并用ELISA双抗体夹心法进行检测 。The Eriocheir sinensis (ES) that suffered 'tremble diease' by artificial infection were homogenated. The supernatant was concentrated with polyethyleneglycol (PEG) 6000 and purified by gel filtration chromatograph with Sepharose 4B. The eluates collected were concentrated at 4℃. The virus particles in the first part of the eluates could be seen under electron microscopy. The diameter of virions varified in 50～100nm. Sinopotamon denticulatum artificial infected with the purified virus became ill. By double antibody sandwich ELISA detection, the results showed that there was significant difference between the infected and the control crabs.
