人睾丸发生过程中TGF β_1表达的研究

摘　　要：研究 TGFβ1 (转化生长因子β1 )在人胚胎睾丸发生过程中的表达。收集 30例 12 - 32周龄胎儿睾丸标本 ,免疫组织化学 ABC法染色 ,以正常羊血清代替一抗为阴性对照 ,TGFβ1 抗血清的工作浓度为 1:10 0 ,光镜观察。结果显示 TGFβ1 在人胚胎睾丸间质细胞呈阳性表达 ,曲细精管内精原细胞、支持细胞、管周肌样细胞未见阳性表达。TGFβ1 在 12周龄睾丸组织未见阳性表达 ,16周～ 32周均有阳性表达 ,在 16周时是表达的高峰 ,此后呈逐渐下降趋势 (P<0 .0 1)。由此结果表明TGFβ1Immunohistochemical techniques of ABC were used to observe the expression of TGF β 1 in 30 testes of human fetus, whose gestational ages were estimated to be 12 to 32 weeks Normal goat serum was used to replace TGF β 1 in the negative control group The positive reaction of TGF β 1 was observed only in Leydig cells In the gestational age of 12 weeks, no positive reaction of TGF β 1 was observed In the gestational ages of 16 to 32 weeks, the positive reaction was present, and the peaks of TGF β 1 expression appeared in the gestational age of 16 weeks With the increase of gestational ages thereafter, the number of positive reaction cells decreased gradully ( P <0 01) This result suggests that TGF β 1 could play an important role during the development of human testis.
