摘 要:为观察凋亡相关基因 - p5 3、bcl- 2在乳腺导管非典型增生及乳腺癌中的表达 ,探讨其与乳腺癌组织发生的关系 ,本实验应用原位杂交方法检测凋亡相关基因 p5 3、 bcl- 2 m RNA ,应用免疫组织化学方法检测 p5 3蛋白在 44例乳腺导管非典型增生组织中的表达 ,并与 6例乳腺导管单纯性增生及 2 6例乳腺癌对比分析。实验结果为 ,p5 3m RNA在乳腺导管单纯性增生组织中呈较强表达 (6 6 .7% ) ,在乳腺导管非典型增生组织中阳性表达为 40 % (轻度 :5 5 .6 % ,中度 :41.7% ,重度 :2 6 .1% ) ,在癌组织中的表达率为 19.2 % (导管内癌 :2 1.4% ,浸润性导管癌 :16 .7% )。p5 3蛋白在导管单纯性增生组无表达 ,在导管非典型增生组阳性表达为 2 4% (轻度 :11.1% ,中度 :2 5 % ,重度 :34 .8% ) ,在癌组织中的阳性表达为 38.5 % (导管内癌 :35 .7% ,浸润性导管癌 :41.7% )。 bcl- 2 m RNA在单纯性增生组无表达 ,在非典型增生组中阳性表达为轻度 :11.1% ,中度 :16 .7% ,重度 :39.1%。在乳腺癌组织中阳性表达为导管内癌 :78.6 % ,浸润性导管癌 83.3%。实验结果表明 ,在乳腺导管重度非典型增生组织中可检测到 p5 3基因有较高的表达缺失、突变及 bcl- 2 mThe aim of the study was to investigate the expression of apoptosis related genes p53?bcl 2 in mammary atypical ductal hyperplasia and breast carcinoma and to explore the relationship between the gene expression of p53?bcl 2 and the progression of breast carcinoma and activity of telomerase In 44 mammary atypical ductal hyperplasia cases,apoptosis related genes p53?bcl 2 were detected by in situ hybridization and expression of p53 protein was detected by immunohistochemistry method,and compared with that of the 6 simple hyperplasia and 26 carcinoma of breast The expression of p53 mRNA was higher in simple hyperplasia(66 7%),40% in atypical ductal hyperplasia(mild 55 6%,medium 41 7%,severe 26 1%)and 19 2% in carcinoma (intraductal carcinoma 21 4%,invasive ductual carcinoma 16 7%) The expression of p53 protein was negative in simple hyperplasia,24% in atypical ductal hyperplasia(mild 11 1%,medium 25%,severe 34 8%),38 5% in carcinoma(intraductal carcinoma 35 7%,invasive ductal carcinoma 41 7%) The expression of bcl 2 mRNA was negative in simple hyperplasia,11 1% in mild, 16 7% in medium and 39 1% in severe atypical ductal hyperplasia, 78 6% in intraductal carcinoma and 83 3% in invasive ductal carcinoma The results showed that the loss of expression and mutation of p53 gene and the excessive expression of bcl 2 mRNA were detected in severe atypical ductal hyperplasia.
