摘 要:用免疫组织化学 SPTM试剂盒标记 ,用 Quantimet5 2 0型图像分析仪定量分析 B波段紫外线照射对 NIH3T3细胞bcl- 2、 n- m yc、 c- fos三种基因表达的影响。结果发现 Bcl- 2 (bcl- 2表达的蛋白质 )定位于细胞质、 N - m yc、 c- Fos (分别为 n-m yc、c- fos表达的蛋白质 )在细胞核中高表达 ,在细胞质中低表达。三种基因蛋白在对照组 (未受紫外线照射的正常 NIH3T3细胞中 )均有表达。NIH3T3细胞经 B波段紫外线照射后 ,9小时前 Bcl- 2无显著变化 ,9小时后急剧增高 ,超过最初值 ;N-m yc于照射后 1小时前无显著变化 ,1小时后降低 ,9小时后显剧增高 ,并超过最初值 ;c- Fos于照射后即下降 ,0 .5小时后回升 。Using immunohistochemical SP TM kits, effects of UVB irradiation on the expression of bcl 2、n myc、c fos in NIH3T3 cell were detected. Then measure their proteins quantitatively with Quantimet 520. It was found that Bcl 2 was located in cytoplasm. Expression of N myc、c Fos in nucleus were more higher than in cytoplasm. Three proteins were all expressed in control sample (normal NIH3T3 cells not irradiated by UVB). Bcl 2 had no apparent change when culture time was less than 9 hours after irradiation, elevated and exceeded initial value markedly after this time. N myc had no apparent change less than 1 hour after irradiation, decreased after this time, elevated and exceeded initial value evidently more than 9 hour after irradiation. c Fos decreased immediately after UVB irradiation, increased 0 5 hour after irradiation and worked up to initial value.\;
