摘 要:检测间隙连接蛋白 Cx43、神经组织蛋白 S- 10 0在去卵巢致骨质疏松症 (OVX- OP)大鼠腺垂体滤泡星形细胞 (FS细胞 )中的表达。实验采用 10月龄未孕产 SD雌性大鼠 40只 ,随机均分为卵巢切除组 (OVX组 ,n=2 0 )和假性手术对照组 (Sham组 ,n=2 0 )。于术后 6周末用双能 X线骨吸收测量法 (DEXA)测量大鼠全身及腰椎 4- 6 (L4- 6 )骨密度 (BMD)。取两组大鼠垂体 ,制成连续切片。应用 FITC标记的 Ig G探针 ,对腺垂体组织中 Cx43和 S- 10 0进行间接免疫荧光染色 ,并用激光扫描共聚焦显微镜 (L SCM)定位和定量分析腺垂体 FS细胞中 Cx43、 S- 10 0的表达。结果发现 ,术后 6周末 OVX组大鼠全身及 L4- 6 BMD均明显低于 Sham组值 (P<0 .0 1,P<0 .0 1)。Cx43阳性荧光反应主要定位于相邻的 FS细胞的胞浆中和 /或胞膜上。OVX组 Cx43阳性表达荧光强度和表达阳性率均显著低于 Sham组 (P<0 .0 1)。S- 10 0蛋白表达定位于 FS细胞的胞浆中 ,两组间 S- 10 0阳性表达荧光强度和表达阳性率无显著差异 (P>0 .0 5 )。本研究提示 ,SD大鼠 OVX术后 6周出现骨质疏松变化 ;OVX大鼠腺垂体 FS细胞数量无明显变化、而 Cx43表达显著下降 ,后者的变化可能与大鼠 OVX-To investigate the expression of gap junction protein connexin 43 (Cx43) and nerve tissue protein S 100(S 100) in adenohypophyseal folliculo stellate cells (FSCs) in the ovariectomized osteoporosis (OVX OP), osteoporosis models of rats were obtained by bilateral ovariectomy. 40 female rats were equally devided into an ovariectomized group (OVX group) and asham operation control group (sham group). Total skeletal bone mineral density (BMD) and spinal(L 4-6 )BMD of each rat were measured with dual energy X ray absorptiometry (DEXA) at the end of 6 weeks after operation. The expression of Cx43 and S 100 in adenohypophysis was detected by immunofluorescent histochemistry and laser scanning confocal microscopy (LSCM). The results showed that the total skeletal BMD and L 4-6 BMD of rats in the OVX group were significantly lower than those in the sham group ( P <0 01, P <0 01). The fluorescence of Cx43 distributed in the cytoplasm and along the cell membrane of FSCs. Both Cx43 fluorescence intensity and Cx43 positive cell ratio were lower in the OVX group than the in Sham group ( P <0 01), while the fluorescence and positive rate of S 100 of FSCs in the OVX group had no discrepancy compared with sham group. These results indicated that the reduction of Cx43 expression in FSCs after ovariectomy could presumably play a role in ovariectomy induced osteoporosis.
