广东省脑膜炎奈瑟氏菌porA基因限制性片段长度多态性分型研究

摘　　要：目的　分析广东省在不同流行时期 ,特别是 80年代后期以来脑膜炎奈瑟氏菌 (Nm)的特点。方法　选择 35株A、B、C群流脑菌株 (196 6～ 2 0 0 0年 )进行porA基因聚合酶链反应 -限制性片段长度多态性 (PCR -RFLP)分型研究 ,采用PCR反应扩增porA基因 ,PCR产物纯化后经限制性内切酶MspI酶切 ;10 %聚丙烯酰胺凝胶 (PAGE)电泳分离酶切产物 ,银染显色。结果　 35株被检菌株分为5种RFLP型 ,2 0株A群菌株 (病人 17株 ,带菌者 3株 )分 3型 ,谱型与我国代表谱型相同 (即a1、a2 、a3型 ) ;11株B群菌株分 5型 ,其中 6株病人菌株每两株 1型 ,分为 3型 (即b2 0 、b2 1、b2 1a2 型 ) ,其中b2 0 型与a3 型谱型相同 ;4株C群菌株有 3株与b2 1a2 菌型相同。结论　不同的血清群可有相同的RFLP谱型 ;A群Nm不同的流行周期有自己优势菌型 ,广东省 6 0年代与 80年代后期至今的代表菌型相同 ,为a3 型 ;B群RFLP谱型比A群变异程度大 ,同一时期分离到的菌株可有几种不同的RFLP谱型 ;Objective To determine the characteristics of Neisseria meningitides (Nm) in Guangdong Province in different epidemic periods, especially after late 1980s. Methods A total number of 35 Nm strains of serogroups A, B, and C isolated from 1966 to 2000 in Guangdong Province were typed by porA genes amplified with PCR, digested with restriction endonuclease Msp I, seperated on 10% polyacrylamide gels and stained with silver. Results\ 35 Nm strains were divided into 5 distinct patterns. 20 strains of serogroup A were divided into 3 distinct patterns (a 1, a 2 and a 3), similar to the predominant of China. 11 strains of serogroup B were divided into 5 distinct patterns, of which 6 strains presented 3 patterns (b 20 , b 21 , b 21 a 2). 3 of 4 strains of serogroup C had b 21 a 2 pattern. Conclusion Nm strains of different serogroups can have the same RFLP patterns. In different epidemic periods, serogroup A was predominant. In 1960s and from the late 1980s to 2000, Strains of a 3 were the representative. Nm strains of serogroup B had several different RFLP patterns, and were more variable than that of serogroup A. Nm strains in Guangdong Province showed a changing trend from serogroup A to serogroup B.
